5. Principle
Nitrofuran (AMOZ) rapid test strips are based on the principle of competitive inhibition immunochromatography. During the flow process, nitrofuran (AMOZ) in the sample binds to nitrofuran (AMOZ)-specific colloidal gold-labeled monoclonal antibody, which inhibits the conjugation of the antibody to nitrofuran (AMOZ)-BSA on the NC membrane detection line combination of things. combination, resulting in a change in the color test line. When the sample has no nitrofuran (AMOZ) residue or the concentration is lower than the detection limit, the T line is darker than the C line or the T line is the same color as the C line; when the concentration is equal to or higher than the detection limit, the T line is obviously lighter than the C line or the T line Lines have no color. The C line appears regardless of the presence or absence of nitrofuran (AMOZ) residues in the sample, indicating that the test is valid.
6. Sample preparation
6.1 Solution Preparation
0.1MK2HPO4 solution: Dilute 1M K2HPO4 solution 1:9 with deionized
water.
6.2 Sample Handling
Tissue:
1) Take 3±0.05g of homogenized tissue sample into a 50ml centrifuge
tube, add 6ml of deionized water, 750ul of 1M HCl and 300ul of
derivatization reagent, shake well for 2min, and take a 60°C water
bath for 30min;
2) Add 7.5ml 0.1M K2HPO4 solution, 600ul 1M NaOH and 10ml ethyl
acetate, shake well for 1min, and centrifuge at 4000r/min for
10min;
3) Take 5ml of the supernatant in a glass tube and dry it at 56°C
with nitrogen or air.
4) Add 2ml of n-hexane, shake well to dissolve the dry residue (if
the residue is not completely dissolved, the result may be
unstable), then add 0.6ml of sample buffer, shake well for 30s,
stabilize until layered, then absorb the upper layer (if it sees
milky white). liquid, centrifuge at 4000r/min for 5min);
5) Aspirate 200ul of the lower liquid (water layer) and prepare for
testing. (Avoid absorbing upper liquid).
7. Operation procedures
7.1 Read the instruction carefully before use. Return test
dipsticks and sample into room temperature.
7.2 Take bottles needed from the kit package, take out required
microwells and dipsticks, making proper marks. Please use these
test dipsticks within 1h. Seal the cap of the bottles, avoid
moisture.
7.3 Take 200ul of the test samples into the microwells, then
repeatedly absorb for 5-10 times, mix the sample with the reagent
in the microwells completely until no solid judged by eyes (this is
a very important step).
7.4 Incubate for 5min at room temperature (20-25℃), insert the test
dipsticks into the microwells with the "MAX" end fully dipped in to
the mixture solution.
7.5 Incubate for 5-8min at room temperature again, read the result,
it is invalid in other time.
8. Test Result Interpretation
8.1 Negative: T line is darker than C line, or T line has same
color with C line. It means there is no Nitrofuran(AMOZ) residue in
sample or the residue is lower than detection limit.
8.2 Positive: T line is obviously lighter than C line or T line is
invisible. It means the Nitrofuran(AMOZ) residue is equal to or
higher than detection limit.
8.3 Invalidation: C line isn’t seen wine red. It means the test
card is out of efficacy, out of date or improper operation. Please
run the test again using another package. If the invalid tests keep
happening, please contact the supplier.
9. Precautions
1) The test card can only be used once at room temperature, please
do not use the expired test card.
2) Do not reuse tips to avoid cross contamination.
3) Do not touch the white film surface in the middle of the test
card, avoid direct sunlight and direct blowing of fans.
4) Wear gloves when testing.

10. Specificity
No cross-reactivity with nitrofurans AOZ, AHD and SEM.
11. Storage and expiration date
Storage: 2-8 ℃ dark, sealed, dry place, not frozen.
